A protocol for CRISPR-mediated activation and repression of human endogenous retroviruses in human pluripotent stem cells double-check efficiency of competent cells used. Agilent XL10 Gold competent cells can be used instead of Stbl3 competent cells for the sgRNA cloning. Perform a BfuA1 digestion on the ligated sgRNA product prior to High Efficiency Transformation for NEB. Stable Competent. E. coli. (C3040H) Thaw a tube of NEB Stable Competent E. coli cells on ice for 10 minutes. Add 1 -2 µl containing 100 pg - 100 ng of plasmid DNA to the cell mixture. Carefully flick the tube 4-5 times to mix cells and DNA. Do not vortex. Place the mixture on ice for 30 minutes. ElectroMAX™ Stbl4™ Competent Cells are specifically designed for cloning unstable inserts. As electrocompetent cells, they have one of the highest transformation efficiencies available (>5×10 9 cfu/µg), making them ideal for generating cDNA and genomic libraries and for cloning unstable inserts. These electrocompetent E. coli: The Mix & Go! transformation kit simplifies competent cell preparation to an easy 3-step protocol that takes only 45 minutes. Any E. coli strain (DH5a, BL21, HB101, JM109, TOP10, XL-1 Blue, XL10 Gold, Zymo10B/DH10B, TG1, ccdB survival, Stbl2, Stbl3, and more…) can be prepared as highly competent cells. Streak out frozen glycerol stock of bacterial cells (Top10, DH5α, etc.) onto an LB plate (no antibiotics since these cells do not have a plasmid in them).Work sterile. Grow plate overnight at 37°C. Standard Transformation Protocol. Transfer the required number of tubes from -70° C freezer to wet ice. Include an extra tube for control DNA, if desired; Allow the cells to thaw for 5 minutes. Gently tap the tubes multiple times to obtain uniform suspension. For control: Add 1 µL (10 ng) pUC19 control DNA to one tube. Alternatives: Although we used the DH5α strain, we recommend either Stbl3 (Thermo C737303) or NEB Stable E. coli strain As this protocol works for me, I would like to know if other people have problems transforming Stbl3 with Gibson products. There is an incompatibility between the Gibson assembly buffer and that of the Stbl3 cells such that the Stbl3 cells are not able to take up your plasmid and therefore all die under antibiotic selection (if you talk to The Stbl3™ E. coli strain is derived from the HB101 E. coli strain and is recommended for use when cloning unstable inserts such as lentiviral DNA containing direct repeats Protocol Search for Additional Bacterial Strains The Stbl3™ E. coli strain is derived from the HB101 E. col
